is it possible to block a room one day after the checkout ? Similarily, to prevent same-day booking, and these settings being exported to all OTA.

Yes it is possible, please go to Settings > Properties > Rooms > Setup and scroll down to Block Dates after Check-out, you can find more details when you use (?) context help.
